DEVELOPMENT OF siRNA DELIVERY SYSTEM FOR THE TREATMENT OF OSTEOPOROSIS

 

Osteoporosis is a skeletal disorder associated with low bone mass and elevation in bone fragility and susceptibility to fractures. It occurs either elderly or postmenopausal or due to endocrine disorders, usage of anticoagulants or receiving chemotherapy. In healthy people, the bone formation and resorption is in balance, however, osteoporosis occurs when this balance is disrupted in the favor of osteoclasts in terms of bone resorption. Its pathogenesis involves two important mechanisms; RANK and M-CSF. siRNA is dsRNA which is used to inhibit the gene of interest’s translation into proteins. In order to overcome the challenges during siRNA delivery to body such as enzymatic degradation, lower efficiency of crossing biological membranes due to negative charge and the need to escape from endosome/lysosome compartments in the cell, siRNA must be delivered with a carrier. Those carrier systems may vary according to the target site. In this study an injectable siRNA-delivery system will be developed for the treatment of osteoporosis.
